What's Meclozine?

Meclozine is actually just a medication used to cure nausea, vomiting and diziness caused by stuff like vertigo and motion sickness.

"meclozine increased body length of Fgfr3ach mice by 5.4% in our study. If the same effect could be observed in patients with ACH, the patients could be expected to increase 6.7–7.1 cm in height"


Note: 5% on a 5'9 individual would be 6'0!

"No radical treatments for short stature in ACH are currently available." we don't know yet but 25mg is usually used when u have motion sickness so just try that prob

FGFR3 modified models (achondroplasia) is NOT the same as normal human growth physiology in normal models H1 receptors inhib doesnt stimulate chondrocyte growth at a relevant level

Meclozine doesnt work for height

explain this then: In contrast, loss-of-function mutations in FGFR3 lead to camptodactyly, tall stature, and hearing loss syndrome

Note: The people with tall stature due to loss of function in FGFR3 never had achondroplasia to begin with

Thats based on a genetic distinction, LOF mutations lower FGFR3s normal inhibitory signals which is the reason for unchecked chondrocyte proliferation and thats what causes the height growth, meclozines effect is from targeting an excessive amount of FGFR3 activity (like i said b4 achondroplasia) and not base FGFR3 signaling which in a normal person who already has balanced FGFR3 (great majority of people) blocking it with meclo has no target, so it doesnt cause the height growth effect u grt with genetic FGFR3 issues

this is kind of a double edged sword, using anti-histamine's also down regulates the adrenal cortex, therby inhibiting a fuckton of enzymes like 21-Hydroxylase (and therby it's products - i.e mineralocorticoids). If you've ever actually used any anti- histamines you would also know that they turn you into a literal narcoleptic, emotionless zombie. Interesting study but really not feasible in practice.

Interestingly, meclozine also increased bone growth in wild-type mice. The plasma concentration ofmeclozine during treatment was within the range that has been used in clinical settings for motionsickness. Increased longitudinal bone growth in Fgfr3ach mice by oral administration of meclozinein a growth period suggests potential clinical feasibility of meclozine for the improvement of shortstature in ACH.

Rats have higher plate plasticity than us and their chondrocytes are more sensitive to FGF3 cause of their receptor activation thresholds and postnatal skeletal growth windows so the increase in bone length isnt as simple to say itll work the same for humans since our chondrocyte are modulated by gh, igf, and thryoid

In the study u sent it says meclozine modultes the FGFR3-ERK axis
It modulating that means it weakens ERK phosphorylation in chondrocytes
The issue with that is ERK signaling in the plates work at a biphasic model and transient activation boosts proliferation while maintained activation inhibits it thru cyclin D1 mediated p21/p27 stabilization
This wont work in humans
Humans ERK signaling have tissue specific crosstalk with RAS-MAPK and PI3K-AKT paths which causes nonlinear feedback loops thatll js stop this from happening
