IGF-1 signaling interacts closely with insulin receptor (IR) pathways due to high structural homology, where both ligands can bind to each other’s receptors to activate shared downstream cascades. Both activate tyrosine kinase receptors that recruit IRS (insulin receptor substrates) and Shc, regulating cell growth, metabolism, and proliferation.
Key Aspects of Interaction and Signaling:

• Receptor Cross-Binding: While IGF-1 binds to the IGF-1 Receptor (IGF1R), it can activate the insulin receptor with lower affinity, and high insulin concentrations can trigger the IGF1R. Hybrid receptors (consisting of one IR and one IGF1R subunit) also exist, binding both ligands.
