Ritalin (methylphenidate) works by blocking the dopamine (DAT) and norepinephrine (NET) transporters, increasing the amount of both neurotransmitters in the synapse. It primarily acts as a reuptake inhibitor, improving attention, focus, and executive function.
Vyvanse/Elvanse (lisdexamfetamine) works differently. It is a prodrug that is converted into dextroamphetamine in the body. Dextroamphetamine not only blocks dopamine and norepinephrine reuptake but also promotes their release from presynaptic neurons, generally producing a stronger, smoother, and longer-lasting effect than methylphenidate.
In simple terms, Ritalin mainly prevents dopamine and norepinephrine from being cleared from the synapse, whereas Vyvanse both prevents their reuptake and actively increases their release. This is why Vyvanse typically lasts around 10–14 hours, while immediate-release Ritalin usually lasts about 3–5 hours.
