This study tested whether berberine (a plant compound known to improve blood sugar) could help prevent muscle loss. Instead, researchers found the opposite.
- Main finding: Berberine actually causes muscle atrophy (muscle loss) in both normal and diabetic mice.
- It does this in two major ways:
- Decreases protein synthesis (muscle building)
- Increases protein degradation (muscle breakdown)
- The key driver is a protein called atrogin-1, which:
- Speeds up muscle protein breakdown
- Also reduces protein synthesis by lowering another important factor (eIF3-f)
- Berberine also:
- Disrupts energy metabolism in muscle (via mitochondria)
- Activates AMPK, which further increases atrogin-1 and suppresses muscle growth pathways
