Anastrozole (Arimidex) and letrozole (Femara) work by reversibly blocking this enzyme, while exemestane (Aromasin) binds to the enzyme and inactivates it permanently.
So is it very bad that my enzyme is inactivated permanently?

I have never heard an aromatase inhibitor may have a permanent effect

Your body makes new aromatase enzymes all the time. Exemestane acts as a pseudosubstrate for the enzyme so it has no function anymore. Once you stop taking it, you go back to normal
