First of all, DHT BLOCKERS ARE THE BEST AT FIGHTING HAIRLOSS! But if you don't want to take them or can't take them then this thread is for you.
Plenty of people are scared of 5ARIs , I am currently not taking them due to developmental reasons, but theres a great alternative especially for young people with aggressive Androgenetic Alopecia.
What is the stack? Its very simple,
DIY long release Minoxidil
KX-826 (Pyrilutamide)
(Ketaconazole as an extra but this is only if your scalp is inflammed and won't be talked about in this thread)
Wtf is "DIY long release Minoxidil"?
Most of you know normal plain minoxidil, either in Topical Formulation or Oral formulation, I personally take it through the oral path due to minoxidil's lethality for cats which I want to avoid.
And some of you may have heard of VDPHL01 , a new in development long form release variant of Minoxidil that is Safer and more Effective (More Hairs per cm²)
This mainly works by blunting Cmax, so the peak concentration of minoxidil in the blood stream, so its low enough to avoid cardiovascular effects and high enough to saturate the Sulfotransferases in the scalp and activate Minoxidil into its active form (Remember that Minoxidil is a prodrug, its not an active drug)
So whilest that drug is still in Phase 3 Trials how can we do a little DIY version of this? Very easy.
1 Insulin Syringe,
1 Topical Minoxidil bottle (5% minoxidil)
1 Liter Water bottle or 0.7l water bottle (MARK THESE BOTTLES IN SOhat iME WAY AND HIDE THEM!! YOU DON'T WANT TO ACCIDENTALLY DOSE YOURESELF OR SOMEONE ELSE WITH 5MG OF MINOXIDIL THAT IS RAPIDLY ABSORBED!)
(The difference of diluting it in different volumes of water is the sip frequency of the bottle, so if you have a 1 Liter bottle you would sip it more often than a 0.7l water bottle to get a similiar Blood concentration curve of minoxidil, I personally prefer 0.7l as you only have to take a big sip every 40 minutes or so to empty the bottle slowly.
make sure that the Minoxidil you buy only has these ingredients : Propylene Glycol , Ethanol , Aqua (Water, H2O) , Minoxidil
THE PROCESS :
Extract 0.1ml of the 5% Minoxidil Solution and put it into your Water Bottle. Thats it. Close both of the containers so the Minoxidil Bottle and the Water bottle. Put the Minoxidil away. Shake your water bottle. And you're done, as Ethanol is water soluable I would say you don't have to shake it everytime before you drink but I just do it regardless.
Make sure you have enough space and that you do this at a sink, you do NOT want to knock the open Minoxidil Bottle over as this would not only be a waste of MONEY but also a VERY MAJOR RISK to any of your PETS and extra unnecessary work.
Now you have a 5mg of Minoxidil diluted in a 1 Liter or 0.7 liter or anything inbetween bottle of water.
You have the Hair Growth stimulant (DIY Long release Minoxidil) Done.
Further down I will tell you HOW to drink it ideally and avoid the usual cardiovascular effects of Minoxidil.
SOME INFO ON KX-826
KX-826 (Pyrilutamide) is a topical anti androgen that is applied locally to the scalp, there it binds to the androgen receptor and competes with DHT, it has a higher binding affinity for the androgen receptor than other known anti-androgens like RU58841 and has gone through 2 Phase 3 Trials now not only proving statistical significance in Hair count improvements over the placebo, but also safety.
Recent Phase 3 Trials showed only a minor difference between the 1.0% concentration group applying KX-826 twice daily to their scalp, and the 0.5% concentration group applying KX-826 twice daily to their scalp. This shows that 0.5% twice Daily is already enough to nearly fully saturate all Androgen Receptors on the areas where its applied, sadly no studies were done on only ones a day application so we don't know how well KX-826 saturates the androgen receptors when only applied ones a day, we also only know that its systemic half life is very short so its hard to estimate.
Twice daily 1.0% KX-826 increased hair count by 15.33 hairs/cm²
Twice daily 0.5% KX-826 increased hair count by 14.46 hairs/cm²
Thats only a relative difference in hair count increase by 6.0% whilest the relative concentration difference is 200.0% (KX-826 0.5% vs 1% concentration)
Looking at this data I would suggest the cheaper lower concentration of 0.5% KX-826 twice a day.
The moment KX-826 goes systemic it quickly gets metabolized into a inactive metabolite of KX-826 called KX-982.
KX-982 has a extremly low binding affinity to androgen receptors CAN NOT bind to androgen receptors.
This prevents systemic Anti-Androgenic effects.
KX-826 will attempt to fight the core issue of Androgenetic Alopecia which is DHT by competing with DHT at the Androgen Receptor, this in combination with its quick metabolization into its harmless metabolite KX-982 prevents and systemic hormonal effects completely.
Here is a before and after after 2 months of treatment (anecdote)
HOW DO WE APPLY THESE NOW?
Your Minoxidil Water bottle will simply be slowly sipped over the course of the day, this can be 8 hours, 10 hours, 12 hours, 14 hours, it doesn't matter that much but your ng/ml concentration of Minoxidil will be constantly high enough to cause hair growth and low enough to avoid any cardiovascular side effects like tachycardia, and most importantly pericardial effusion.
We will be applying the KX-826 twice a day on the areas where we are balding, this mean we will apply it ones in the morning and ones before bed.
KX-826 is very lightweight and usually absorbs pretty quickly so it wont leave a greasy residue.
With this stack you will FOR SURE regrow hair and maintain.
Im excited to talk about GT20029 in the future which is another drug in development currently by the same people who developed KX-826 and works in a very unique way.
If you want exact math for ng/ml blood concentration of minoxidil with mathematical formulas so you can be exact in your protocol and get the desired effects and avoid side effects then utilize this below.
Bottle Volume (in ml) / Sip Volume = Total Sips , assuming average larger sips we will go with 40ml sip volume. So youd input.
Bottle Volume (in ml) / 40ml = Total Sips for a 0.7l bottle this is 17.5 sips.
For dosage you need
Total Dosage (in mg) / Total Sips
Since we are doing 5mg of minoxidil (you can also experiment with higher and lower mg ammounts) I will input 5mg into total dosage and since I will be taking 17.5 sips throughout the day to empty my 0.7l bottle I will input 17.5 Sips , this will look like this
5 (mg) / 17.5 (sips) = 0.28mg per sip .
To get the time between each sip we will need this formula
Time (in minutes) / (Sips - 1) = Time between each Sip (in minutes)
(The -1 in (Sips -1) is there because we will have 16.5 intervals between the beginning sip and ending sip so we ignore the one before the first sip.)
So for me this would mean im gonna be sipping this for around 10 hours so 600 minutes, and I have a total of 17.5 sips so I input
600 (Minutes) / (17.5 (sips) - 1) = 36.3 (Minutes)
So according to the math I am ingesting 0.28mg per 1 sip every 36.3 minutes, and after 17.5 sips im finished with my 0.7l bottle.
To get a graph of your ng/ml I would have to write a computer script that runs a specific formula and calculates every single sip which I'm not gonna do right now.
But with the protocol im doing in the calculations my Cmax (peak concentration of Minoxidil in the blood stream) should be around 10.5ng/ml which is well below the cardiovascular threshold and above the sulfotransferase threshold, so again low enough to avoid the cardiovascular side effects (occur at around 20ng/ml) and high enough to continously feed the sulfotransferase enzyme in the scalp with minoxidil (which starts at around 2ng/ml).
