Firstly, it is well known that BMP2 increases RUNX2 expression and, consequently, osteoblast differentiation.

The mechanism of action of WAY262611:
Firstly, DKK1 is inhibited. When DKK1 is inhibited, the suppression of the Wnt signalling pathway is reduced. As Wnt signalling increases, β-catenin accumulates in the cell. β-catenin enters the cell and binds to TCF/LEF. Once this complex binds to specific regions of the DNA, BMP2 transcription occurs.

BMP2 transcription in detail:
The information on how to synthesise the BMP2 protein is already present in the nucleus. β-catenin enters the nucleus. A β-catenin+TCF/LEF complex forms in the nucleus. TCF/LEF recognises specific sequences on the DNA and transcribes the information from the BMP2 gene onto RNA. Subsequently, the mRNA (messenger RNA) transmits this information to the ribosome. The ribosome reads the mRNA and functions as follows: ‘I must produce the BMP2 protein according to the instructions in this mRNA’.

Looking at the mechanism here, theoretically, WAY262611 could influence bone formation as it facilitates the synthesis of the BMP2 protein. However, if anyone knows the basis for the claim that it locally promotes the growth of facial bones, please do share your insight.
