Researchers find that they can successfully estimate the population of non-motile organisms by using quadrat sampling even though it presents challenges. This method places a square frame (quadrat) in systematic locations within the study area. The species' percentage cover or the number of organisms is recorded within each quadrat and is maintained.

Once researchers collect the data and average it, they can subsequently scale it up in order to estimate the total population residing in the area. This method works best for sessile organisms like plants and algae and fungi because they remain stationary. This method reliably assesses population density and distribution and frequency, while it minimizes biases.

You can see an example here, where along the transects they made quadrats of 2m and then recorded the species they found.

P.S. Make sure the quadrats are at a random place in the study area, otherwise there can be biases even if it's subconcious.
