In today’s video I’ll be speaking about another theory I have though of and is combining saline with localised bone growth agents to stimulate controlled apposition growth in your desired area this can also work for the audience who’s growth plates are open as a lot of the apposition growth compounds normally are net negative for growth plates etc .
Ok so the main dissolving agent we will be using is saline, saline is used medically to mimic cosmetic results of an implant or facial surgery of ur choice it lasts 6-12 h normally so the rate at what it dissolves is slow and steady making perfect for a slow release of growth factor ans ideally the growth factors should be water soluble as saline has been used with many water soluble compounds even in the medical field.
Ok so now that we got the what is saline out the way we will talk about the compounds we can use and mix in saline in theory to cause localised bone growth could dissolve super slow and over time causing appositional growth in ur desired area one of the drugs I’ll be cover today is Rosuvastatin.
Rosuvastatin is a cholesterol medication ( statin ) that have been proven to cause localised bone growth in animal models it does this by up regulating bmp2 and increasing the number of osteoblast in the area where it’s taking effect thus resulting in localised appositional growth and not by a small amount by about 30-50% increase yes 30-50% that’s a crazy amount as u can even see in the study : ( I’ll put them at the end )
So why do statins do this how do they increse bmp2 . It does this because stains Inhbit the HMG-CoA reductase enzyme. Normally this enzyme is responsible for converting HMG-CoA into mevalonate thus making the cell experience a shortage of non sterol lipid intermediates ( the most significant ones in this case are FPP and GGPP ) normally these lipids act as a negative break on ostoblast by keeping bone building genes off .
When FFP and GGPP are spurred by stains the ras and RhoA can’t be prenylated this means they remain trapped inside the cytosol of the cell not being able to to keep catabolic enzymes like mkp-1 ( catabolic protein that normally puts a break to cell growth signals ) thus meaning that bc of the mkp-1 down regulation the growth and anabolic path ways of the cells can’t be down regulated and shut down. ( this is down via RhoA)
Because also stains stop ras as well this causes the cell to panic and this causes a rapid raise in cytosolix ras his hyper active ras now can bind to the enzyme called PI3K and the PI3K then converts membrane lipids into messenger that can then up regulate and active Akt what then increases then increases of the cell and and blocks apoptosis in the osteoblast forcing them to lay down new bone and matrix did longer .
Simultaneously the over active raf protein also phosphorylates mek1/2 what then their goal is to locate and bind to the erk1/2 and phosphorylates then as well what the allows it to travel to the nucleus of the cell and have its anabolic genomic effects like massively up regulating runX2 osterix and AP-1 this then massively up regulates bmp2 and the cells then secrete bmp2 locally what then binds to any local stem cells forcing them to turn unit hard bone matrix
Another anabolic that can be used is magnesium chloride when applied locally it dissolves into magnesium ions and then enter MSCs what this then causes the up regulation of notch1 what then tells those MSCs to mature into ostoblast to start depositing bone thus triggering appositional and local bone growth while also the magnesium ions trigger sensory nerves in the periosteum to release and neurotransmitter called CHRP what then boosts the production the Periosrin a matrix protein crucial in Cortial widening and appositional growth while also up regulating anabolic genes like runX2 and other anabolic path ways as seen in this graph
Also I want to mention that all these compounds would have superb synergy with the already know anabolics for bone like abalo Romo etc since they up regulate anabolic bone genes potentially causing even more growth and faster results .
