Linoleic acid and its effects on periosteal apposition and increse in Cortial thickness without affecting Endocortial bone.
LA ( linoleic acid ) it’s a polyunsaturated fat typically naturally found in sunflower/ other seed oils yes seed oils
.
Now the question is how would this fat help us achieve a more dimorphic / robust skeletal structure thru appositional growth ?
LA when injected subq in rats at 0.3mg per kg ( for humans I believe that would be 0.05mg per kg rough estimate ) has been shown to increase appositional bone formation by up to 220% and increse Cortial thickness by over 38% while also having 0 effect on Endocortial bone .
This means all the bone growth that was observed in the rats was true bone expansion and not just the marrow of the bone getting tighter and more narrow as most “ bone mass “ drugs normally do so there was an actual structural change and it was even noted that there was an increase in periosteal perimeter by 15% .
Ok so how does LA do this it does this by being an agonist to the PPAR-alpha/delta receptors. When we activate these receptors as in the study with LA this caused a sharp spike in ostoecalcin and a huge increase in the number of local osteoprogenitor cells to turn into mature osteoblasts also turning on the ppra-a/d also tells local stem cells in the periosteum to to aggressively differentiate into osteoblasts hence why we can see that massive spike in bone formation rate in the periosteum another use for the ppra-a is that activating it optimised how the cell processes fatty acids for energy/atp .
