Downstream affects of those genes is what causes UC, we dont fully understand the pathways so its too risky, knowing what the problem is isnt enough, also editing one gene wont fix the entire network

oh i skimmed through it ive read this b4, the only correlation it has is to IBD but if ETS2 influences macrophages in UC, then it could be a partial solution, the study was more about finding drugs supressing ETS2 without broad immune supression, tbh never reached anything further than that, its just a theory that they didnt prove nor disprove

Just tests MEK inhibitors its not the same as the other study, MEK inhibitors arent safe long term, maybe if they found a delivery method to target macrophages it could work, ETS2 is more a path they can use to find treatments not one thatll be a solution

First restore the microbiome with precission probiotics or any bacteriophage that specifically eliminate only harmful bacteria and boosts useful species like Faecalibacterium and Akkermansia since they promote metabolites like butyrate to repair the gut lining, next step is probably strengrhening the epithelial barrier using small molecules like MitoQ or any compound that increases ATP production and stabilizes tight junction proteins, immune mods can be done using biologics like IL-23 or TNF inhibitors, combining that with JAK inhibitors to suppress local inflam without immunen supression widespread, epigentic reprogram could be done using HDAC inhibitors, which already are in trial phases for autoimmune, and then lastly ud need an advanced drug delivery systems, like pH sensitive nanoparticles (i have a guide on my post that had a DIY nanoparticle guide i wrote in it) or an oral forumla that somehow would release meds only into the colon

Why would anyone want surgery as a first option? It’s incredibly invasive and presents risks. If you could treat the condition and improve quality of life of medication beforehand, then that is logically the better choice.

But anyways, have fun suffering eating food and not knowing whether or not you’ll be on the toilet in pain for the days after, all because you’re too pussy to get the surgery you need

What immunosuppressant did you use? Biologics target more specific pathways rather than general suppression
 
I think you used it as eye drops and I think it's a local suppressant

No I don’t take it anymore. My condition improved and went away as we tapered down the dosage until I was fine without it. I don’t use any treatments anymore.

I occasionally get flare ups but they go away without intervention most of the time. If I get a flare up that is really bad, which hasn’t happened in a long time, I will use a Dexamethasone eye drop that will stop the inflammatory pathway locally and return my eyes to normal.

This was about 4 years ago
