Dopamine is an essential neurotransmitter that greatly benefits us in terms of survival through its effect on motivation. However, it also has effects on testosterone and growth hormones.
Low to moderate dopamine = BOOST IN TEST AND GROWTH HORMONES
Excessive dopamine = NUKE TEST AND GROWTH HORMONES
Therefore, boosting dopamine WITHOUT inhibiting GnRH neurons is essential. Here, in this study, rats were given a dopamine agonist and an antagonist to compare how it influences GnRH and whether it boosts testosterone levels. They also measured how DHT affects it, but that's not important right now. What they found in the study is this -
PMID: 1612019
In another study, treatment with Mucuna pruriens also helped to restore androgens and even fix the HPTA, but only in infertile men, still showing the importance of dopamine -
PMID: 18973898
THEREFORE - Ensure dopamine is maxxed out. Remove stress. Occasional nicotine, caffeine and sugar are actually beneficial to spike dopamine and reduce cortisol short term. It's the desensitisation to the receptors that nukes it, the oxidative stress damaging leydig cells, and cortisol increase over time.
Take mucuna idk

Ahhhh, the relaxing neurotransmitter. That also boosts GROWTH HORMONE!!!!!!

Valproate (GABA transaminase inhibitor = ↑ GABA) blunted GH response by ~50% at all time points, and so GABA seems to be inhibitory for GH overall????
The first assumed pathway is this:
Diazepam → GABA-A activation → Dopamine release ↑ → GH ↑
However, in a different study, there was found to be a different verdict.
PMID: 6841564
In this study, when diazepam (which boosts GABA activity) was combined with L-dopa and apomorphine (which increase dopamine), it reduced the amount of growth hormone (GH) released.
This suggests that GABA can limit GH release by affecting dopamine. Normally, dopamine helps stop GH release, and by boosting GABA, diazepam seems to block dopamine's effect, making it harder for dopamine to stop GH from being released.
So the pathway is now this -
Diazepam → GABA-A activation → Reduction in Dopamine activity → Less dopamine inhibiting GH → GH ↑
The increase in GH from diazepam seems to be linked to GABA reducing dopamine’s inhibition on GH release, making it easier for GH to be secreted.
In humans, acute oral GABA administration elevates plasma growth hormone levels, while prolonged treatment blunts growth hormone response and enhances prolactin response to insulin induced hypoglycemia (Cavagnini et al., 1980)
Basically, over prolonged use of GABA, instead of releasing GH during hypoglycemic metabolic states (during fasting, after exercise etc), the balance shifts to more prolactin being released
Avoid daily use or cycle the GABA
okay i give up ill finish later

LOCATIONS WHERE DOPAMINE HITS
Hypothalamus
Dopamine INHIBITS GnRH neurons (less LH, less testosterone) ---------------- (PMID: 23239814)
Pituitary
Suppression of prolactin through D2 receptors, which increases testosterone and GH/IGF-1 due to less suppression of GnRH/LH
Testes
Dopamine stimulates Leydig cells, which increases steroidogenesis and therefore T - EVEN WITHOUT A SURGE IN LH

"BRO", the agonist bromocriptine, increased GnRH mRNA in certain brain cells (neurons) by 67%, showing dopamine's importance in increasing testosterone and androgen levels.a 14-day treatment with BRO increased by 67% the number of silver grains per neuron

Serum T and LH levels, as well as seminal plasma and blood levels of dopamine, adrenaline, and noradrenaline were also decreased in all groups of infertile men. This was accompanied by significantly increased serum FSH and PRL levels in oligozoospermic subjects. Treatment with M. pruriens significantly improved T, LH, dopamine, adrenaline, and noradrenaline levels in infertile men and reduced levels of FSH and PRL

This boost in GH is actually from dopamine, as they found in the study that the use of pimozide (basically a dopamine blocker that crosses the brain blood barrier) blunts the spike in GH, and this boost is not affected whatsoever by prolactin. In 9 normal volunteers, GABA caused a significant increase of plasma GH levels (P < 0.0001 vs. GH levels in a group of 14 controls)
PMID: 370137
Despite the increase after a benzodiazepene (diazepam), blocking dopamine with pimozide cuts this GH spike by 50%, showing that it is again involved in the process of GABA boosting GH.mean serum GH level increased 336% 1 h after diazepam administration
