AMD3100 (blocks CXCR4)


Mirabegron (β3-adrenergic agonist)


Dislodge MSCs from bone marrow into the blood, then direct them to growth plates with mechanical loading.

AMD3100 (Plerixafor)


AMD3100 is a CXCR4 antagonist.
It blocks the CXCL12 (SDF-1) / CXCR4 axis, which normally keeps stem cells anchored in the bone marrow niche.
This leads to mobilization of stem cells into the bloodstream.

Multiple studies on hematopoietic and mesenchymal stem cell mobilization.


Mirabegron (ß3-Agonist)


Mirabegron is a selective ß3-adrenergic receptor agonist.
It can help dislodge MSCs from their niche and increase mobilization when combined with AMD3100.


Mirabegron + AMD3100 Synergy


The combination significantly increases the number of circulating mesenchymal stem cells (MSCs) compared to either drug alone.
These MSCs can potentially be directed to specific areas with mechanical stress (e.g- joint loading or plyometrics).

Theoretical Height Application


If growth plates still have cartilage left (not fully ossified):
Mobilized MSCs could potentially migrate to the growth plate area and differentiate into new chondrocytes → more cartilage stacking → possible continued growth.


Major Limitations & Warnings:


MSCs don't automatically go to growth plates.


They may not integrate properly or form new functional cartilage.


No human trials for height growth exist.


Both drugs have side effects and require medical supervision.



Current Research Status

The combination has been studied mainly for:

Cardiac repair after heart attack
General MSC mobilization
Other regenerative applications
Not for epiphyseal plate regeneration or height increase.


Role of Mechanical Loading


The suggestion is to combine drug-induced MSC mobilization
with targeted mechanical stress (e-g-lateral joint loading or plyometrics) to
guide the cells to the desired area.


This is still theoretical and unproven for height growth.


Conclusion


Mirabegron + AMD3100 is an interesting pharmacological approach to mobilize stem cells.
Whether it can be used for meaningful height increase in humans remains unknown and requires proper clinical research.
