ASP-4345
ASP-4345 is a research Positive Allosteric Modulator the of the Dopamine D1 Subtype which are mostly located in the Prefrontal Cortex, which is the most abundant excitatory (cAMP increasing) dopamine receptor in the brain. To simplify it will boost your natural response to dopamine, making you more sensitive without directly acting as a agonist and attaching directly to the receptor and activating it. That means to reap the effects you must engage in a task for the natural agonists to release and their effect be amplified. In my personal experience this is best paired up with other dopamine-increasing compounds.
-improved working memory, better signal to noise ratio, better executive function.
GB-115
GB-115 is a Anxiolytic that has also been found to have cognitive enhancing properties that is available in Russia as a prescription under the name ranquilon. It's primary mechanism works as a Cholecystokinin Receptor Antagonist, having a higher affinity for CCK2 and moderate affinity for CCK1. These receptors can be found in fear processing regions like the amygdala, hippocampus, and cortex. They have been shown to induce anxiogenic Effects and panic attacks. It is also a mild BRS3 Receptor antagonist which has been associated with higher levels of anxiety. It has been shown to lower cortisol levels and can even be used as a sleep aid if you struggle with nighttime anxiety. Ideally it should be dosed twice a day due to its short half life (effects linger on for way longer), Both nasal and oral are fine but you are gonna irritate your nasal mucosa with spray. One of my favorites in my personal experience and I would recommend for anyone looking to relief anxiety long term in a safe and pro cognitive way, as a lot of anxiolytics also dumb you down.
- Anxiolytic, Cognitive Boost, Improved Mood and less tress, Anti depressant
Coluracetam
Has a pretty unique mechanism with it primarily functioning as high-affinity choline uptake (HACU) enhancer. This is the tightly regulated rate limiting step in the synthesis of acetylcholine directly dictating how much acetylcholine a neuron can manufacture. This in leads to a greater amount of acetylcholine available which controls attention, learning, memory, and arousal. This should be paired up with a Choline source, preferably CPD Choline for best results.
- enhanced visual perception, improved memory and focus, Neuroprotection, Cognitive Flexibility
Guanfacine
Guanfacine is a secondary choice non-stimulant ADHD medication that works in a completely different mechanism than most stims. It functions as a selective alpha2A-adrenergic receptor agonist on dendritic spines in the prefrontal cortex. Guanfacine strengthens the PFC signal and this is part of the brain regulates focus, working memory, executive function, and high level thinking. This specific subtype is an inhibitory receptor that reduces intracellular cAMP (Too much is bad), calming the brain down. It is notable for acting as a break for norepinephrine triggering a negative feedback loop that tells nerve cells to release less norepinephrine, which leads to its blood pressure and heart rate lowering effects. Promotes the maturation and stability of dendritic spines more specifically mushroom Spines which hold strong long-term memories and are associated with long-term synaptic stability. You will need a prescription for this one.
- Less ADHD like behavior (Reduced Hyperactivity, Anxiety, impulsivity), Improved Executive function, Better signal to noise ratio, working memory
