EXPLANATION:
PTH analogs are a synthetic, injectable version of of the parathyroid hormone. Parathyroid hormone works on increasing the calcium level in the blood, by releasing calcium directly from bone deposits. While it may sound counterintuitive, injecting parathyroid intermittently actually INDUCES OSTEOBLAST ACTIVITIY. When PTH levels are continuously high, It's primary objective is breaking down bone tissue. Conversely, when the body is exposed to PTH in short, intermittent bursts, the opposite effect occurs. These brief pulses stimulate bone-building cells, leading to an increase in bone formation. This anabolic effect is the principle that underlies the therapeutic use of PTH analogs, which mimic these short pulses to build bone.
USES:
The main medical application for PTH analogs is the treatment of severe osteoporosis, particularly in individuals who are at a high risk for fractures. These medications are prescribed for postmenopausal women or men who have not had a sufficient response to other osteoporosis therapies or are intolerant to them. They are also approved for treating osteoporosis associated with the long term use of systemic glucocorticoid medications. Another use is to manage hypocalcemia in patients with hypoparathyroidism. So what does this actually mean for you? Hypothetically, it could increase dimorphism, open room for more appositional growth, however it doesn't fix recession. These PTH analogs can induce bone formation at ANY age, as the room for appositional growth is open extremely long periods of life.
MECHANISM OF ACTION:
PTH analogs function by promoting the formation of new bone, achieved by stimulating osteoblasts when injected intermittently. the analog binds to PTH receptors on osteoblasts, triggering signaling pathways that promote their survival and enhance their bonebuilding work.
PROTOCOL:
Cumulative use of PTH analogs will actually cause more harm than good, as the effects disappear and begin to wane after a period of time (usually 18-24 months).
The most common injection sites are the thigh or the abdominal region, preferably for your first few doses you're gonna wanna pin where you can sit in case you get a headache or get dizzy. The types of PTH analogs you're gonna wanna pin are: Abaloparatide & Teriparatide.
EXTRAS:
Bare in mind, there aren't many in this community that have gone through with this, and it should be the absolute last resort before surgery. It increases overall bone mass, and you may end up with disappointing results depending on how sensitive your facial receptors are. Do not try to inject this locally.
