I randomly discovered this compound while looking into direct stem cell therapies on growth plates, which I might turn into a separate post if I find it interesting enough.
Smoothened Agonist (SAG) stimulates Hedgehog (Hh) signaling, which promotes the activity and proliferation of stem cells in the growth plate, leading to increased bone length rather than premature closure like other remedies involving Hh. Rats and mice with locally implanted beads containing SAG or intra-articular injections increased the proliferation of epiphyseal skeletal stem cells in the growth plate, leading to lengthened limbs compared to controls.
So theoretically, SAG can be used as a topical / local injection to prevent growth plate closure and increase limb length in humans. However, the compound is only used for professional research sourced exclusively through authorized suppliers.
