Matrixyl doesn't work.
Matrixyl is a peptide in a huge number of serums and gets cited constantly. The evidence is almost nothing.
There's one real study. Robinson 2005. 93 women, 12 weeks. All six authors worked at Procter & Gamble. It used 3 parts per million of the peptide. That's the entire clinical case for original Matrixyl.
Everything else is marketing. The "collagen +105%" numbers, the fibroblast data, the Synthe'6 trial — that's all Sederma's own unpublished brochure material. Retailers host it as PDFs and people link it like it's a paper.
The doses you see quoted aren't the peptide. Synthe'6 as sold is 0.025% actual peptide in glycerin and water (it's in patent US9149659). So a "2% Synthe'6" serum contains 0.0005% peptide. Five ppm. Matrixyl 3000 is the same trick. And those percentages aren't proven doses — they're the supplier's suggested use range.
Cagefuel: In cell culture, you need roughly 100–500 nM to make a fibroblast do anything. At 3 ppm topically, you'd only hit that if 100% of it crossed the skin barrier intact. Choi 2014 measured it and found barely any reaches the dermis, and what does get in breaks down fast. Real-world you're 100–1000x below the level that does anything in a dish
One actual warning: don't dermastamp it. Park 2017 found pal-KTTKS at the concentrations needling actually achieves pushes fibroblasts toward the scarring phenotype (CTGF and α-SMA up, myofibroblast conversion). The authors flagged it themselves. The only reason topical Matrixyl is harmless is that nothing gets through — needling removes that. Also don't stamp in any non-sterile serum, that's how people get facial granulomas.
tl;dr: one industry study at 3 ppm, the "2%" on the label is 99.9% glycerin, nothing reaches the dermis, no one has ever found more collagen in treated skin. Harmless, useless, overpriced. Buy Tretinoin.
