Tideglusib is a compound which no one has really talked about yet however holds promise for bone growth and is something which everyone needs to consider.

Tideglusib is an irreversible inhibitor of Gsk3 with phase 2 trials and is the most potent inhibitor we have available.

GSK-3β (glycogen synthase kinase 3 beta) is a key regulator of bone growth, primarily through its role in the Wnt/β-catenin signalling pathway. GSK-3β phosphorylates β-catenin, targeting it for degradation, which reduces osteoblast activity and slows bone formation. When gsk-3b is inhibited, β-catenin increases and activates genes that promote osteoblast proliferation, differentiation, and mineralization, leading to increased bone formation. B catenin also inhibits osteoclastogenesis indirectly. Basically you want high b catenin for bone growth and inhibiting gsk3b which marks b catenin for degradation is the best way to do that.

Hyperactive gsk-3b is also implicated in depression, bipolar disorder and AD; gsk-3b is a negative regulator of synaptic plasticity and impairs LTP promoting cell death. Gsk-3b also promotes pro inflammatory cytokines and reduces glycogen storage

Tldr: gsk3b is a shit enzyme and needs to be nuked so tideglusib shows promise

'Just take lithium bro jfl' Yes lithium is a inhibitor of gsk3b however at the higher doses necessary to produce maximal inhibition it is toxic to thyroid and also raises PTH which is not what we want.
