Selegiline is an irreversible MAO-B inhibitor that remains highly selective for the MAO-B enzyme at standard oral doses below 10 mg per day. Because it binds irreversibly, the enzyme it attaches to is essentially taken out of commission until the body synthesizes new MAO-B — a process that generally takes 10–14 days. MAO-B’s main job in the brain is to break down dopamine. When selegiline blocks it, less dopamine is metabolized and more dopamine remains available. By preventing dopamine from being broken down, selegiline effectively enhances dopaminergic signaling in the brain. With more dopamine remaining available in synapses for longer, dopaminergic neurons can maintain stronger and more sustained communication. This boost in dopamine tone supports motor function and other dopamine-dependent processes without directly forcing dopamine release. It works purely by preserving the dopamine that’s already there.

When dopamine is broken down by MAO-B, the process produces several potentially toxic by-products, including hydrogen peroxide and DOPAL. These oxidative metabolites can contribute to oxidative stress and cellular damage, especially in dopamine-rich areas of the brain. By inhibiting MAO-B, selegiline not only keeps dopamine around longer but also reduces the formation of these oxidative by-products, which is one of the reasons it has been explored for possible neuroprotective effects.

Because selegiline increases the availability of dopamine, many of its effects in humans stem from this enhanced dopaminergic signalling. In clinical contexts, supporting dopamine pathways can help improve motivation, initiative, mood, and cognitive drive, since dopamine plays a key role in reward, goal-directed behaviour, and mental energy. Some individuals also report improvements in libido or overall interest and engagement in daily activities, which aligns with dopamine’s involvement in pleasure, desire, and reinforcement.

With doses below 10mg orally per day it remains largely selective for mao B inhibiting nearly 100% of the enzyme at this dose. Some minor MAO A inhibition may occur at this dose but not irreversibly and only by a small amount. At doses below 10mg daily tyramine diet restriction is not required but may be wise to avoid food sources exceptionally high in tyramine. Combining selgelline and stimulants should be approached with caution as selegelline will potentiate any dopaminergic drug even nicotine or caffeine. If your taking other substances please ensure its safe to combine with selegeline. Also other dosing routes such as sublingual or transdermal can result in higher bioavailability meaning loss of selectivity for MAO-B could occur at lower doses. Let me know what you guys think.
