Apitegromab (SRK-015) | The Most Promising Myostatin Inhibitor To Date
Apitegromab (SRK-015) is an experimental myostatin inhibitor currently under development. It is administered intravenously (IV). Scientific/medical studies have definitively proven that apitegromab has a significant effect on preserving and even increasing lean muscle mass, with almost zero risks or side effects. In a study published in Nature, the subjects taking apitegromab + tirzepatide (Mounjaro/Zepbound) preserved 54.9% more lean muscle mass relative to the group taking tirzepatide + placebo only; in absolute terms, apitegromab preserved 1.9 kg (4.2 lbs) more lean muscle mass relative to the placebo group.

Apitegromab has passed Phase 1, Phase 2, and Phase 3 trials, and is currently awaiting final approval by the Food and Drug Administration (FDA). The target date by which the FDA will make a decision is September 30, 2026.

What is apitegromab?
Apitegromab is a fully human monoclonal antibody that inhibits precursor forms of myostatin; specifically, it works to stop the activation of promyostatin and latent myostatin. By preventing these precursors from activating, it successfully reduces active myostatin signaling levels in the body and promotes muscle preservation and growth. It was developed by Scholar Rock to treat spinal muscular atrophy (SMA) by targeting muscle weakness.

Myostatin is a protein that regulates and inhibits muscle growth. It is naturally produced by the body. Studies on animals and humans have shown that organisms who have reduced production of myostatin exhibit muscle growth well beyond the levels of the average organism.
Any side effects or risks?
No, not really. No serious side effects have been reported in trials and studies. A trial/study published in Advances in Therapy, a peer-reviewed medical journal, concluded "Apitegromab was safe and well tolerated, on the basis of the adverse event (AE) profile with no clinically meaningful changes in baseline vital signs, electrocardiograms, or clinical laboratory parameters and no anti-drug antibody formation.

Another trial/study published in Frontiers in Neurology concluded "The benefit of apitegromab treatment observed at 12 months was sustained at 36 months with no new safety findings."
