Basically melanotan 2 agonizes MC1R and then it produces a tan. Question is, would one cycle of melanotan be enough to permanently alter the state of a gingers MC1R and cause them to have a different skin pigmentation all together?

No. Why would anyone permanently change after one "cycle". I must have used a hundred vials within the last 10 years and I would still eventually go completely white. A ginger friend of mine used it to get his beard dark and he actually got vitiligo from using it (permanent white patches).
