Sodium valproate
What is sodium valproate?
Dekepane, Valcote
is a medication for people with epilepsy that induces differential bone growth through long-term epigenetic changes.
It inhibits HDACs (Histone Deacetylases), which cause the activation of genes related to bone growth.
RUNX2 – master transcription factor for osteoblast differentiation.
BMP2 – Bone Morphogenetic Protein 2, promotes bone formation.
ALP – Alkaline Phosphatase, early marker of osteogenesis.
COL1A1 – Collagen Type I Alpha 1, essential structural protein in bone.
OPN – Osteopontin, bone matrix protein.
BSP – Bone Sialoprotein, involved in bone mineralization.
Cons: It has severe short- and long-term liver damage.
(Acetylcysteine can help prevent this.)
At doses higher than 2000 mg can be fatal, cause death, coma,
It can cause hair, eyebrow, and eyelash loss if the dose is exceeded
Low testosterone with a high dose
Pros: It can inhibit the gene responsible for aromatase CYP19A1
It can be obtained without a prescription
Increases chondrocyte proliferation
By inhibiting HDACs, genes such as SOX9 and IGF1 are activated, which stimulates chondrocytes to divide more, expanding the growth plate.
Promotes chondrogenic differentiation
Improves chondrocyte maturation to a stage where they can be replaced by bone through endochondral ossification (a key process for bone lengthening).
Increases extracellular matrix production
Activates genes such as ACAN and COL2A1, which code for proteins such as aggrecan and type II collagen, essential for growth plate.
Prevents premature chondrocyte senescence
This means that chondrocytes do not "age" as quickly, which prolongs their activity in the growth plate.
Regulates apoptosis (programmed cell death)
It can reduce unnecessary chondrocyte death, helping maintain a healthy, growing chondrocyte population.
It enhances the IGF-1 effect.
If you're going to use it, use it without exceeding 1000 mg
You should use it every day
It has a shelf life of 9 to 16 hours after ingestion
I hope it helps.

Valproic acid prevents glucocorticoid‑induced osteonecrosis of the femoral head of rats
Glucocorticoids (GCs) are the most common cause of atraumatic osteonecrosis of the femoral head (ONFH) because their effect compromises the osteogenic capability of bone marrow‑derived mesenchymal stem cells (BMSCs). Valproic acid (VPA) is a widely used anti‑epileptic and anti‑convulsant drug...

Histone deacetylase inhibition with valproic acid downregulates osteocalcin gene expression in human dental pulp stem cells and osteoblasts: evidence for HDAC2 involvement - PubMed
Adult mesenchymal stem cells, such as dental pulp stem cells, are of great interest for cell-based tissue engineering strategies because they can differentiate into a variety of tissue-specific cells, above all, into osteoblasts. In recent years, epigenetic studies on stem cells have indicated...
