Not even a minor thing either, he stated that E2 should not be lowered because it messes with Collagen X, RUNX2, and SOX9 when that's the goal..? He made it seem like it was a bad thing. Clearly GPT slop. No one here knows anything. EVERYTHING is based upon the PTHrP loop, stop studying things such as the wnt/b-catenin pathways when you don't know the prerequisites.

Bud was talking about wnt/b-catenin causing the cells to go from proliferative -> pre hypertrophic regardless of estrogen levels when SOX9 doesn't allow for that to happen (SOX9 is higher with lower E2 if you couldn't tell). The argument has 0 merit since he claims lowering E2 is irrelevant for height.

What would work is upping the PTHrP levels which in turn ups SOX9 levels. This makes the cells move down the growth plate slower while also replicating more. Chondrocytes primarily end due to low PTHrP levels which is due to going further down in the growth plate as the PTHrP levels are a gradient

Imagine the pinker it is, the higher the PTHrP levels. The closer you get to the left side (the bone), the lower the levels are.

SOX9 is the main controller of cells going from the proliferative phase -> pre hypertrophic, the wnt/b-catenin pathways are lower in power. It directly stops other pathways from interfering if they try assuming the SOX9 levels are high enough.

So, what's the solution..? Waiting for AI chips to send signals to the pituitary gland to secrete GH will likely be done before all this.. It would be very difficult to get studies done on this anyways due to the risks.

Locally injecting PTHrP can somewhat help if done properly as it could get into the growth plate but it's not reliable. There probably are some pathways that you could manipulate but it's going to get really twisted.
