Recently, I've been seeing a lot of iqlets coping with Erdafitinib, thinking it's somehow going to change their height or make them taller. This seems to come from the assumption that FGFR3 inhibition alone will promote growth, while ignoring the fact that Erdafitinib is a pan-FGFR inhibitor with significant effects on multiple FGFR pathways.
Erdafitinib inhibits FGFR2, which directly governs growth plate biology. Silencing FGFR2IIIc narrows the proliferative and hypertrophic chondrocyte zones, reduces IHH and PTHrP expression, delays ossification, and suppresses RUNX2/SPP1 all essential for longitudinal growth. FGFR3 inhibition in isolation could theoretically be growth promoting, but the simultaneous FGFR2 damage far outweighs any benefit and Erdafitinib even inhibits fgfr4, wich can cause severe eye damages. So probably the worst fgfr3
